The BENCE JONES TEST is an innovative diagnostic kit designed for the determination of kappa and lambda light chains (Bence Jones proteins) in urine, using the double immunodiffusion method. This test is essential for the diagnosis of conditions such as multiple myeloma and other conditions associated with high production of these proteins.
Product Features
- Composition: Each plate of the BENCE JONES TEST consists of a layer of agarose gel, with sodium azide (1 g/l) as a preservative.

- Storage: Store at temperatures between +2° and +8°C. Usable until the expiration date indicated on the label.
Test Principle
The test is based on the Ouchterlony technique, which allows the determination of kappa and lambda light chains through immunodiffusion in agarose gel. The antigens and antisera, deposited in different wells of the gel, diffuse and react forming a visible band of precipitate, indicating the presence of these proteins in urine.
Execution Procedure
- Open the plate and let it sit at room temperature for about 5 minutes to allow any water droplets to evaporate.
- Inoculate 40µl of the urine sample into the central well.
- Add 10µl of kappa antiserum into the right side well and 10µl of lambda antiserum into the left side well.
- Let absorb for 20 minutes, then close the plate with the lid and invert it.
- Store the plate in its pouch and leave it at 37°C for 9-15 hours for diffusion.
Result Evaluation
Result evaluation is simple and straightforward:
- Absence of bands: Excludes the presence of kappa or lambda light chains in urine.
- Presence of bands: Indicates the presence of light chains; the position of the band allows estimating the concentration of the sample.
The sensitivity of the method is approximately 2.8 mg/dl for K chains and 2.1 mg/dl for lambda chains.

- 1 Tube of Anti h-Ig/kappa chains x 400 µl
- 1 Tube of Anti h-Ig/lambda chains x 400 µl
- 1 Tube of Positive control type kappa and lambda x 100µl
Warnings and Precautions
It is recommended not to ingest the product and to avoid contact with skin and mucous membranes, especially when using in vitro diagnostics containing sodium azide.
