Method Principle
The Contact Slide 2 uses two types of media:
- Rose Bengal CAF Agar + Neutralizing: This medium is enriched with Rose Bengal and Chloramphenicol, which inhibit bacterial growth and limit the size of rapidly growing mold colonies. The rose bengal stains yeast and mold colonies pink, facilitating identification.
- PCA + TTC + Neutralizing: Contains triphenyl tetrazolium chloride as a growth indicator, forming an easily observable insoluble red compound.
Testing Procedure
Follow these simple steps for effective sampling:
- Take a slide from the refrigerator and allow it to acclimatize to room temperature for about 5 minutes.
- Unscrew and remove the slide from the cylindrical container, avoiding contact with the agar surface.
For surface monitoring:
- Bend the cap to form a 90° angle and press each side of the slide firmly against the surface to be examined for 10 seconds.
- Alternatively, use a swab to sample the area and gently roll the swab on the agar surface.
- For liquid examination, hold the slide by the cap and immerse it completely in the test fluid.
- Reinsert the slide into the test tube, screw it in, and incubate at 30 ± 1 °C for up to 5 days. Record the count on PCA + TTC + Neutralizing after 72 hours of incubation.
Interpreting Results
Count the total number of colonies on PCA + TTC + Neutralizing (side 2) to obtain the total bacterial count. The total number of colonies grown on Rose Bengal CAF Agar + Neutralizing (side 1) provides an assessment of fungal contamination.
