DIF-STAIN Composition
The DIF-STAIN dye consists of:

Staining Protocol
Protocol for Smears
- Air dry the smear and fix it in the DIF-STAIN Fixative.
- Pour off excess Fixative.
- Immerse the slide 5 times for 1 second in Solution A, removing excess liquid after each immersion.
- Repeat the previous step with Solution B.
- Shake the smear to mix the dyes.
- Wash in spring water and rinse in distilled water or Weise buffer pH 7.2.
- Allow the slide to dry in an upright position.
Protocol for Paraffin Sections
- Bring the sections to water.
- Stain with Solution A for 30 seconds.
- Stain with Solution B for 30 seconds.
- Quickly rinse in distilled water.
- Dehydrate and clarify.
- Mount the slide and observe under the microscope.
Interpretation of Results
The DIF-STAIN staining allows for a detailed color scheme, useful for interpreting samples. The results can be compared with those of the classic May Grünwald – Giemsa method:
- Eosinophilic Granules: Reddish-brown
- Basophilic Granules: Blue
- Nuclei: Blue-violet
- Nuclei of Blood Parasites and Protozoa: Bright red
- Neutrophilic Granules: Red-violet
- Lymphocyte Protoplasm: Blue with possible reddish-purple granules
- Platelets: Blue with a violet central body
- Erythrocytes: Pale pink
- Microorganisms: Blue-violet
- Spermatozoa: Dark blue
