General Description:
URIN SYSTEM Plus is an advanced diagnostic system designed for the determination of total microbial load, the identification of microorganisms, and the performance of the antibiogram directly from urine samples. The system uses a panel of 24 wells containing dried biochemical substrates and antibiotics. After inoculation with a dilution of urine and incubation at 36 ± 1 °C for 18-24 hours, the results are interpreted through the color change of the wells.

Method Principle:
URIN SYSTEM Plus allows the simultaneous execution of total microbial load, microbial identification, and antibiogram directly from urine samples in 18-24 hours.

• 
  Total Microbial Load: Evaluated through microbial growth in wells 1-GR+ and 2-GR++, containing growth indicators.
  


• 
  Microbial Identification: Based on growth characteristics in wells 3-ESC to 9-CAN, which contain substrates for differential culture.
  


• 
  Antibiogram: Evaluated on the basis of growth or inhibition of microorganisms in wells 10-AK to 23-SXT, with well 24-C serving as a control.
  

Sample Collection and Storage:

• 
  Collection: Urine must be collected aseptically in a sterile container, preferably the first morning urine or at least 6 hours after the last urination. Samples should be sent to the laboratory immediately after collection. If they cannot be processed right away, store them in a refrigerator between 2 and 8 °C for no more than 24 hours.
  


• 
  System Storage: Store at 2-8 °C in the original packaging until the expiration date indicated on the label.
  

Test Procedure:

1. 
   Sample Preparation: Perform microscopic examination of the urinary sediment to verify the presence of microorganisms. Transfer 0.5 mL of urine into a vial of Physiological Solution (Suspension A). Then transfer 0.2 mL of Suspension A into a vial of Suspension Medium (Suspension B).
   


2. 
   System Inoculation: Using single-channel or multichannel pipettes, inoculate wells 1-GR+ to 9-CAN with 0.2 mL of Suspension A for microbial load and identification. Inoculate wells 10-AK to 24-C with 0.2 mL of Suspension B for the antibiogram.
   


3. 
   Incubation: Cover the system and incubate at 36 ± 1 °C for 18-24 hours.
   


4. 
   Result Interpretation: Observe the color change of the wells to interpret the results using the table provided in the instructions. The control well (24-C) must be positive; if negative, repeat the test.
   

Quality Control:
Each batch of URIN SYSTEM Plus is tested with reference bacterial strains to ensure the quality and reliability of the results.

Performance and Reliability:
The results obtained with URIN SYSTEM Plus are in line with those obtained with traditional methods, such as URITEST N dip slides and radial diffusion tests according to Bauer et al. This system shows greater effectiveness in the isolation of Enterococcus spp. compared with traditional dip slide methods.
